Evaluation of the chick as a test animal in the assay of diphtheria toxoids.

چکیده
In a previous report (Branham and Wormald, 1954), the authors have described the titration of therapeutic diphtheria antitoxins in 7to 8-day old chicks. These antitoxins had a titer of several thousand units per ml. Reference was made to an adaptation of this technique to the determination of smaller amounts of antitoxin in the sera of guinea pigs vaccinated with precipitated toxoids. An effective means of titrating small amounts of antitoxin is especially desirable because of the widespread use of toxoids for active immunization of children and the trend towards vaccination of adults. The recommended guinea pig test (National Institutes of Health, 1947) is sometimes inconvenient to perform inasmuch as suitable guinea pigs are expensive and at times are difficult to obtain. Different strains of guinea pigs vary in susceptibility to diphtheria toxin; sex too is a factor of considerable importance (Kolb et al., 1955), and variation among individual pigs also occurs. The reliability of chicks in the studies already described suggested further investigation of their adaptability to the evaluation of prophylactic diphtheria toxoids.

Humoral immune response to Diphtheria and Tetanus toxoids by intranasal administration
The immunogenicity of ten different formulations of intranasal diphtheria and tetanus vaccines which containing different absorption enhancers, adjuvants and other excipients were determined in guinea pigs by the serum neutralization (SN) method. From these ten formulations, it was selected four formulations which gave significant immunogenicity in guinea pigs. In order to design the "final f...
متن کاملIncorporation of T-cell epitopes from tetanus and diphtheria toxoids into in-silico-designed hypoallergenic vaccine may enhance the protective immune response against allergens
Objective(s): New generation of allergy vaccines is capable of promoting the development of protective IgG and blocking the functionality of allergen-specific IgE. We incorporated universal and powerful T-cell epitopes from tetanus and diphtheria toxoids (TD epitope) into recombinant Che a 2, the well-known allergic profilin of Chenopodium album, to determine its immun...
متن کاملPreparation and Evaluation of Diphtheria Toxoid-Containing Microspheres
Preparation of chitosan (CS) microspheres as a novel drug delivery vehicle for intranasal immunization using high, medium and low CS molecular weight (MW) was investigated in this study. Diphtheria toxoid (DT) was used as a model antigen. The emulsion-solidification method was adopted for microencapsulation of DT. In the first step, following the purification of semi-crude DT by the ion-exchan...
متن کاملA New Precision–Fed Chick Assay for Determining True Metabolizable Energy Values of some Poultry Feed Ingredients for Broiler Chickens
Two experiments were conducted to determine: first, the best time of excreta collection after force feeding of broilers to yield the maximal amount of excreta, and second, nitrogen-corrected true metabolizable energy contents of some poultry feedstuffs including yellow corn, soybean meal, corn gluten meal, canola meal and poultry by-product meal by a new precision-fed chick assay using 3-week-o...
